    Competitive Binding Between Id1 and E2F1 to Cdc20 Regulates E2F1 Degradation and Thymidylate Synthase Expression to Promote Esophageal Cancer Chemoresistance

    Purpose: Chemoresistance is a major obstacle in cancer therapy. We found that fluorouracil (5-FU)-resistant esophageal squamous cell carcinoma cell lines, established through exposure to increasing concentrations of 5-FU, showed upregulation of Id1, IGF2, and E2F1. We hypothesized that these genes may play an important role in cancer chemoresistance. Experimental Design: In vitro and in vivo functional assays were performed to study the effects of Id1–E2F1–IGF2 signaling in chemoresistance. Quantitative real-time PCR, Western blotting, immunoprecipitation, chromatin immunoprecipitation, and dual-luciferase reporter assays were used to investigate the molecular mechanisms by which Id1 regulates E2F1 and by which E2F1 regulates IGF2. Clinical specimens, tumor tissue microarray, and Gene Expression Omnibus datasets were used to analyze the correlations between gene expressions and the relationships between expression profiles and patient survival outcomes. Results: Id1 conferred 5-FU chemoresistance through E2F1-dependent induction of thymidylate synthase expression in esophageal cancer cells and tumor xenografts. Mechanistically, Id1 protects E2F1 protein from degradation and increases its expression by binding competitively to Cdc20, whereas E2F1 mediates Id1-induced upregulation of IGF2 by binding directly to the IGF2 promoter and activating its transcription. The expression level of E2F1 was positively correlated with that of Id1 and IGF2 in human cancers. More importantly, concurrent high expression of Id1 and IGF2 was associated with unfavorable patient survival in multiple cancer types. Conclusions: Our findings define an intricate E2F1-dependent mechanism by which Id1 increases thymidylate synthase and IGF2 expressions to promote cancer chemoresistance.     Low mass fraction impregnation with graphene oxide (GO) enhances thermo-physical properties of paraffin for heat storage applications

    Whereas previous researchers analyzed the thermal behavior of paraffin waxes impregnated with graphene oxide nanoparticles (P-GONP) at high mass fraction ( > 1%), this paper analyzes behavior and stability at only 0.3% mass fraction. GONP was prepared by Hummer’s method. The morphology was studied using scanning electron microscope (SEM), transmission electron microscope (TEM), X-Ray diffraction (XRD) and Fourier Transformation-Infrared (FT-IR) Spectrometer and the thermal properties were measured using laser flash analyser (LFA), differential scanning calorimetry (DSC), thermo-gravimetric analysis (TGA) and thermal cycling. LFA showed a 101.2% and 94.5% increase in the thermal conductivity of P-GONP compared to pure paraffin (P) in solid and liquid state respectively. Melting and solidifying temperatures and latent heat were found to be 63.5, 59 °C & 102 kJ/kg and 57.5, 56 °C & 64.7 kJ/kg for P and P-GONP respectively. Thermal cycling over 4000 cycles showed that P-GONP was 27% more stable than P. The latent heat was 64.7 kJ/kg, a 36.5% deterioration compared to virgin paraffin. Compared against higher mass fraction impregnation, lower mass fraction P-GONP was found to have almost equivalent thermo-physical properties (namely thermal conductivity, melting and solidifying characteristics, thermo-chemical stability and reliability) while providing considerable cost saving

    Joint bilateral learning for real-time universal photorealistic style transfer

    Photorealistic style transfer is the task of transferring the artistic style of an image onto a content target, producing a result that is plausibly taken with a camera. Recent approaches, based on deep neural networks, produce impressive results but are either too slow to run at practical resolutions, or still contain objectionable artifacts. We propose a new end-to-end model for photorealistic style transfer that is both fast and inherently generates photorealistic results. The core of our approach is a feed-forward neural network that learns local edge-aware a ne transforms that automatically obey the photorealism constraint. When trained on a diverse set of images and a variety of styles, our model can robustly apply style transfer to an arbitrary pair of input images. Compared to the state of the art, our method produces visually superior results and is three orders of magnitude faster, enabling real- time performance at 4K on a mobile phone. We validate our method with ablation and user studies.Published versio

    A multi-site study on walkability, data sharing and privacy perception using mobile sensing data gathered from the mk-sense platform

    Walking is a fundamental part of a physically active lifestyle, it is one of everyday activities that positively impacts health and wellbeing. In this paper we describe the challenges and experiences of conducting a sensing campaign in the wild. We make use of mk-sense; a software platform to facilitate the deployment of collaborative sensing campaigns. We elaborate on two cross-cultural studies conducted in four different countries (Mexico, Turkey, Spain, and Switzerland) with a total of 77 participants. We present a detailed description of the data collected from one of the studies aimed at measuring walkability around three different university campuses. The analysis of the data shows that walkability can be assessed using information from the sensors in the smartphones and results from surveys answered by participants.
